Just a tip - found this out today and I don't think it has been posted elsewhere.
If you are using the mysql CLI at the same time as SilverStripe is trying to build, you can sometimes cause this issue (usually Windows) - just exit the CLI

That's certainly an unusual cause for that error message; it is MySQL stating a lock issue. I'd expect that sort of concurrancy fault with Microsoft Access but not MySQL...
This error from my experience is generally a problem when your disk has run out of space, or your database is corrupt, or if you have file permission problems. (The latter possibly being the cause of your fault?)
