500 - Internal server error uploading file

I have a fresh installation of SilverStripe on Win Server 2008. Everything seems to be working beautifully EXCEPT that, while logged in as an admin, we cannot upload files. Immediately after clicking on "Upload Files Listed Below", the server throws "500 - Internal server error. There is a problem with the resource you are looking for, and it cannot be displayed."

Any idea what's wrong? Permissions issue?

I had to grant Write and Modify permissions to the local Users group for the assets directory.
