Hey Sam!

i make the folder silverstripe-cache on the root and then i get into admin/assets?flush=1 and works perfectly.

I had the same issue as Juan I think. Just deleted all the files in "/tmp/silverstripe-cache-[mydomainname]/" and that fixed the problem.
